Shoot differentiation in G. abyssinica was observed when callus obtained from hypocotyl transferred to MSO (Murashige and Skoogs medium 1962 without any phytohormone) with various combination of phytohormones. Serial microtome sections through callus grown on MSO containing cytokinins (BAP, KN and zeatin) revealed that callus which appears unorganised externally had internal cellular differentiation. From section of callus of different ages it was noticed that initiation of differentiation of trachiedal element to shoot bud formation takes place within one week to three week of sub culture of callus. Shoot bud formation was clearly exogenous in origin.
